The pH of an organic solution is determined by the molecule's ability to donate or accept protons, which dictates transport across membranes.
Ionization Factors
- Stronger acids readily donate protons, existing primarily in an ionized state.
- Ionization impacts lipid solubility: ionized forms do not cross the blood-brain barrier easily.
- Henderson-Hasselbalch equation describes the relationship between pH and the ratio of ionized to unionized drug forms.
Clinical Application
- Manipulating urinary pH can promote the excretion of drugs by changing their ionization state (e.g., salicylate poisoning).
